Circumventing Antineoplastic Drug Resistance: When Tumor Cells Just Say “No” to Drugs

چکیده

Antineoplastic drug resistance represents a specialized case of drug tolerance since cells most responsive to therapy are destroyed. Clinically, drug resistance may be observed at the onset of therapy or is acquired later by a tumor cell population over time due to a combination of spontaneous genetic mutations and selection by cytotoxic chemotherapy. Numerous mechanisms of drug resistance have been elucidated from in vitro studies but their relevance to clinical drug resistance has only recently begun to be appreciated. Newly described drug resistance mechanisms in tumor cells

A role for survivin in chemoresistance of endothelial cells mediated by VEGF.

Although standard anticancer chemotherapeutic drugs have been designed to inhibit the survival or growth of rapidly dividing tumor cells, it is possible to enhance the efficacy of such drugs by targeting the proliferating host endothelial cells (ECs) of the tumor vasculature. A theoretical advantage of this strategy lies in the possibility of circumventing, or significantly delaying, acquired d...

Circumventing tumor resistance to chemotherapy by nanotechnology.

Patient relapse and metastasis of malignant cells is very common after standard cancer treatment with surgery, radiation, and/or chemotherapy. Chemotherapy, a cornerstone in the development of present day cancer therapy, is one of the most effective and potent strategies to treat malignant tumors. However, the resistance of cancer cells to the drugs remains a significant impediment to successfu...

Novel anti-cancer drug COTI-2 synergizes with therapeutic agents and does not induce resistance or exhibit cross-resistance in human cancer cell lines

Emerging drug-resistance and drug-associated toxicities are two major factors limiting successful cancer therapy. Combinations of chemotherapeutic drugs have been used in the clinic to improve patient outcome. However, cancer cells can acquire resistance to drugs, alone or in combination. Resistant tumors can also exhibit cross-resistance to other chemotherapeutic agents, resulting in sub-optim...

Cross-resistance to Vincristin and Etoposide in a sub line of the human breast cancer T47D cells selected for Adriamycin-resistance

Breast cancer is one of the most common malignancies among women. Although chemotherapy remains a major therapeutic approach to treat cancers, drug therapy often fails for several reasons, particularly the drug resistance. Resistance to multiple chemotherapeutic agents is one of the most important problems in the treatment of different types of cancers. Therefore, in this study a resistant sub ...
